Specimen: 3 mL whole blood in 1 Blue Top (Sodium Citrate) tube. Mix thoroughly by inversion. Transport to Lab within 4 hours. If this is not possible, make PPP within 1 hour of the collection as follows: Centrifuge sample at 3600 rpm for 15 min. & transfer supernatant to a clean plastic tube. Centrifuge this supernatant again at 3600 rpm for 15 mins. & finally, transfer the supernatant (PPP) to 1 labeled clean plastic screw-capped vial. FREEZE IMMEDIATELY. Ship frozen. DO NOT THAW.

Usage: Deficiency of Factor VIII causes prolongation of APTT that corrects completely on mixing. Congenital Factor VIII deficiency is called Hemophilia A which is inherited in an X-linked recessive manner. It affects 1 per 5000 to 10000 live male births. Males are affected whereas females are carriers. The severe form generally presents in infancy with spontaneous bleeding in soft tissues, joints, or intracranial/urinary/gastrointestinal bleeding. Factor VIII is also decreased in moderate & severe forms of von Willebrand disease (VWD).
