Alzheimer’s disease (AD) is a degenerative disease of the brain that causes dementia, gradual loss of memory, judgment, and ability to function. This generally appears in people older than age 65, but less common forms of the disease appear earlier in adulthood. Most cases of early-onset Alzheimer’s disease are caused by gene mutations that can be passed from parent to child. It has been found the disorder can result from mutations in one of three genes: APP, PSEN1, or PSEN2. Another gene APOE has also been associated as a risk factor for the disorder & is a major genetic risk for AD. The presence of 1 allele of this gene increases the risk 2 to 3 fold whereas the presence of 2 alleles increases the risk 16 fold in both sexes. Any mutations of these genes lead to accumulation of large amounts of a toxic protein fragment, amyloid beta-peptide in the brain forming clumps called amyloid plaques, which are characteristic of Alzheimer’s disease. |
