EX0031      ADAMTS13 ACTIVITY
Specimen: 3 mL whole blood in 1 Blue Top (Sodium Citrate) tube. Mix thoroughly by inversion. Transport to Lab within 4 hours. If this is not possible, make PPP within 1 hour of the collection as follows: Centrifuge sample at 3600 rpm for 15 min. & transfer supernatant to a clean plastic tube. Centrifuge this supernatant again at 3600 rpm for 15 mins. & finally, transfer the supernatant (PPP) to 1 labeled clean plastic screw-capped vial. FREEZE IMMEDIATELY. Ship frozen. DO NOT THAW. Overnight fasting is preferred.

Usage: ADAMTS13 (a disintegrin and metalloprotease with thrombospondin type 1 motif, 13) is an enzyme that specifically cleaves usually large von Willebrand Factor (VWF) multimers. If the activity of ADAMTS13 is decreased, either because of an inherited mutation within the ADAMTS13 gene or because of the development of an autoantibody, the accumulation of ultra-large VWF multimers may cause microvascular thrombosis due to platelet aggregation and produce a syndrome of Thrombotic Thrombocytopenic Purpura (TTP). This assay is performed to confirm TTP diagnosis and may also be used in the prediction of TTP relapse.
